    Through studies in mammalian model systems, the estrogen-receptor-related receptor (ERR) alpha, an orphan nuclear receptor, has been shown to interfere with estrogen signaling and might therefore be an interesting pharmaceutical target in estrogen-related diseases. ERRalpha is also involved in energy storage and consumption, and its modulation might be of relevance in the treatment of obesity and diabetes. Recent data have also been published on the effects of this receptor, as well as other members of the ERR family, in non-mammalian animal model systems. Besides indications concerning their mechanisms of action, this analysis demonstrated a role for ERRalpha in controlling cellular movements, and suggested that ERRs might be implicated in a more subtle range of processes than originally envisioned

    Gestion informatisée des connaissances pour une agriculture durable

    International audienceAgriculture must evolve into a more environmentally-friendly way while being economically workable. This type of agriculture is said to be sustainable. It has a systemic logic and therefore requires a lot of knowledge. Our work is based, on the one hand, on inquiries into the knowledge flows in conventional and sustainable agriculture. On the other hand, we investigate a more theoretical approach based on a state of the art of knowledge management applied to the industrial world. These two approaches lead to show the relevance of developing a sustainable agriculture knowledge management tool. Finally, from two methods, we have identified priority knowledge to capitalize in this tool.L'agriculture devra évoluer vers une agriculture plus respectueuse de l'environnement tout en étant économiquement viable. Ce type d'agriculture est dite durable. Elle a une logique systémique et nécessite donc beaucoup de connaissances. Notre travail repose d'une part sur des enquêtes sur la circulation des connaissances en agriculture conventionnelle et durable, et d'autre part sur une approche plus théorique fondée sur un état de l'art de la gestion des connaissances appliquée au monde industriel. Les deux démarches conduisent à montrer la pertinence de développer un outil de gestion des connaissances en agriculture durable. Enfin, à partir de deux méthodes, nous avons identifié les connaissances prioritaires à capitaliser dans cet outil

    Estimation of fungal diversity and identification of major abiotic drivers influencing fungal richness and communities in northern temperate and boreal Quebec forests

    Fungi play important roles in forest ecosystems and understanding fungal diversity is crucial to address essential questions about species conservation and ecosystems management. Changes in fungal diversity can have severe impacts on ecosystem functionality. Unfortunately, little is known about fungal diversity in northern temperate and boreal forests, and we have yet to understand how abiotic variables shape fungal richness and composition. Our objectives were to make an overview of the fungal richness and the community composition in the region and identify their major abiotic drivers. We sampled 262 stands across the northern temperate and boreal Quebec forest located in the region of Abitibi-Témiscamingue, Mauricie, and Haute-Mauricie. At each site, we characterized fungal composition using Illumina sequencing, as well as several potential abiotic drivers (e.g., humus thickness, soil pH, vegetation cover, etc.). We tested effects of abiotic drivers on species richness using generalized linear models, while difference in fungal composition between stands was analyzed with permutational multivariate analysis of variance and beta-diversity partitioning analyses. Fungi from the order Agaricales, Helotiales, and Russulales were the most frequent and sites from the north of Abitibi-Témiscamingue showed the highest OTUs (Operational Taxonomic Unit) richness. Stand age and moss cover were the best predictors of fungal richness. On the other hand, the strongest drivers of fungal community structure were soil pH, average cumulative precipitation, and stand age, although much of community variance was left unexplained in our models. Overall, our regional metacommunity was characterized by high turnover rate, even when rare OTUs were removed. This may indicate strong environmental filtering by several unmeasured abiotic filters, or stronger than expected dispersal limitations in soil fungal communities. Our results show how difficult it can be to predict fungal community assembly even with high replication and efforts to include several biologically relevant explanatory variables

    A coiled-coil affinity-based system for the controlled release of biofunctionalized gold nanoparticles from alginate hydrogels

    Affinity-based systems represent a promising solution to control the delivery of therapeutics using hydrogels. Here, we report a hybrid system that is based on the peptidic E/K coiled coil affinity pair to mediate the release of gold nanoparticles (NPs) from alginate scaffolds. On one hand, the gold NPs were functionalized with the Ecoil-tagged epidermal growth factor (EGF). The bioactivity of the grafted EGF and the bioavailability of the Ecoil moiety were confirmed by EGF receptor phosphorylation assays and by capturing the functionalized NPs on a Kcoil-derivatized surface. On the other hand, alginate chains were modified with azido-homoalanine Kcoil (Aha-Kcoil) by azide–alkyne click chemistry. The hybrid system was formed by dispersing NPs functionalized with the Ecoil-tagged EGF in alginate hydrogels containing either 0, 10, or 20% of Kcoil-modified alginate (Alg-Kcoil). With 20% of Alg-Kcoil, the release of Ecoil-functionalized NPs was reduced by half when compared to the release of NPs without Ecoil, whereas little to no differences were noticed with either 0 or 10% of Alg-Kcoil. Differential dynamic microscopy was used to determine the diffusion coefficient of the NPs, and the results showed a decrease in the diffusion coefficient of Ecoil-functionalized NPs, when compared to bare PEGylated NPs. Altogether, our data demonstrated that the E/K coiled coil system can control the release of NPs in a high Kcoil content alginate gel, opening diverse applications in drug delivery

    Vulnerability of CMOS image sensors in megajoule class laser harsh environment

    CMOS image sensors (CIS) are promising candidates as part of optical imagers for the plasma diagnostics devoted to the study of fusion by inertial confinement. However, the harsh radiative environment of Megajoule Class Lasers threatens the performances of these optical sensors. In this paper, the vulnerability of CIS to the transient and mixed pulsed radiation environment associated with such facilities is investigated during an experiment at the OMEGA facility at the Laboratory for Laser Energetics (LLE), Rochester, NY, USA. The transient and permanent effects of the 14 MeV neutron pulse on CIS are presented. The behavior of the tested CIS shows that active pixel sensors (APS) exhibit a better hardness to this harsh environment than a CCD. A first order extrapolation of the reported results to the higher level of radiation expected for Megajoule Class Laser facilities (Laser Megajoule in France or National Ignition Facility in the USA) shows that temporarily saturated pixels due to transient neutron-induced single event effects will be the major issue for the development of radiation-tolerant plasma diagnostic instruments whereas the permanent degradation of the CIS related to displacement damage or total ionizing dose effects could be reduced by applying well known mitigation techniques

    Boreal forest multifunctionality is promoted by low soil organic matter content and high regional bacterial biodiversity in Northeastern Canada

    Boreal forests provide important ecosystem services, most notably being the mitigation of increasing atmospheric CO2 emissions. Microbial biodiversity, particularly the local diversity of fungi, has been shown to promote multiple functions of the boreal forests of Northeastern China. However, this microbial biodiversity-multifunctionality relationship has yet to be explored in Northeastern Canada, where historical environment have shaped a different regional pool of microbial diversity. This study focuses on the relationship between the soil microbiome and ecosystem multifunctionality, as well as the influence of pH and redox potential (Eh) on the regulation of such relationship. Structural equation modelling (SEM) was used to explore the different causal relationships existing in the studied ecosystems. In a managed part of the Canadian boreal forest, 156 forest polygons were sampled to (1) estimate the α- and β-diversity of fungal and bacterial communities and (2) measure 12 ecosystem functions mainly related to soil nutrient storage and cycling. Both bacteria and fungi influenced ecosystem multifunctionality, but on their own respective functions. Bacterial β-diversity was the most important factor increasing primary productivity and soil microbial biomass, while reducing soil emitted atmospheric CO2. Environmental characteristics, particularly low levels of organic matter in soil, were shown to have the strongest positive impact on boreal ecosystem multifunctionality. Overall, our results were consistent with those obtained in Northeastern China; however, some differences need to be further explored especially considering the history of forest management in Northeastern Canada

    Vulnerability of optical detection systems to megajoule class laser radiative environment

    The Laser MegaJoule (LMJ) facility will host inertial confinement fusion experiments in order to achieve ignition by imploding a Deuterium-Tritium filled microballoon [1]. In this context an X-ray imaging system is necessary to diagnose the core size and the shape of the target in the 10-100 keV band. Such a diagnostic will be composed of two parts: an X-ray optical system and a detection assembly. The survivability of each element of this diagnostic has to be ensured within the mixed pulse consisting of X-rays, gamma rays and 14 MeV neutrons created by fusion reactions. The design of this diagnostic will take into account optics and detectors vulnerability to neutron yield of at least 1016. In this work, we will present the main results of our vulnerability studies and of our hardening-by-system and hardening-by-design studies
